Moderate exercise boosts immune system, diminishes flu's severity in mice

It appears as though exercise pain does have plenty of gain when it comes to fighting off the severe effects of the flu. A new study by five Iowa State University researchers on mice infected with the flu virus suggests that a moderate workout per day may just keep the doctor away -- or at least diminish the severity of the flu's symptoms.

The researchers found that mice that regularly ran on a treadmill over a three-and-a-half month period developed less severe symptoms from the flu virus than those that were not subjected to exercise, and had less influenza virus in their lungs. The mice that regularly exercised showed lower levels of inflammatory factors in their lungs soon after being infected with the virus.

"Perhaps the moderate stress from repeated exposure to moderate exercise might then improve your ability to respond to other stresses, such as influenza," said Marian Kohut, an ISU associate professor of kinesiology and the study's lead researcher. "We're continuing to try and find out if that's true, then what are the mechanisms?"

What's more, even a group of mice that ran on the treadmill for just 45 minutes on the day of their infection experienced some short-term benefit in diminishing the severity of the flu.

"The improvements in host defense resulting from a single session of exercise were quite surprising," Kohut said. "Just exercising one time had some benefit, even though it was more short-term than the mice that exercised regularly. They [those that exercised regularly] did better across the full course of infection. But those mice that exercised just once had some benefits in the first few days of infection."

Gene discovered –p16 that 'cancer-proofs' rodent's cells

Scientists have discovered that the naked mole rat, the only known cancerless animal, has two-tier defense against cancer. Despite a 30-year lifespan that gives ample time for cells to grow cancerous, a small rodent species called a naked mole rat has never been found with tumors of any kind—and now biologists at the University of Rochester think they know why.

 

 

The findings, presented in today's issue of the Proceedings of the National Academy of Sciences, show that the mole rat's cells express a gene called p16 that makes the cells "claustrophobic," stopping the cells' proliferation when too many of them crowd together, cutting off runaway growth before it can start. The effect of p16 is so pronounced that when researchers mutated the cells to induce a tumor, the cells' growth barely changed, whereas regular mouse cells became fully cancerous.

"We think we've found the reason these mole rats don't get cancer, and it's a bit of a surprise," say Vera Gorbunova and Andrei Seluanov, professors of biology at the University of Rochester and lead investigators on the discovery. "It's very early to speculate about the implications, but if the effect of p16 can be simulated in humans we might have a way to halt cancer before it starts."

Like many animals, including humans, the mole rats have a gene called p27 that prevents cellular overcrowding, but the mole rats use another, earlier defense in gene p16. Cancer cells tend to find ways around p27, but mole rats have a double barrier that a cell must overcome before it can grow uncontrollably.

"We believe the additional layer of protection conferred by this two-tiered contact inhibition contributes to the remarkable tumor resistance of the naked mole rat," says Gorbunova in PNAS . Gorbunova and Seluanov are now planning to delve deeper into the mole rat's genetics to see if their cancer resistance might be applicable to humans.

Suboptimal levels of Vitamin D in US children

In previous posts I've stressed the importance of Vitamin D in protecting the body from influenza. Now we see a worrying study that illustrates the suboptimal exposure of  most US children--Latest analysis confirms suboptimal vitamin D levels in millions of US children..National data suggest non-whites are especially at risk

Millions of children in the United States between the ages of 1 and 11 may suffer from suboptimal levels of vitamin D, according to a large nationally representative study published in the November issue of Pediatrics, accompanied by an editorial. (If you check you'll likely find the same defecits in Native Indian Canadian children too--and hence the higher risk of dying from swine flu. Walter Derzko)

The study, led by Jonathan Mansbach, MD, at Children's Hospital Boston, is the most up-to-date analysis of vitamin D levels in U.S. children. It builds on the growing evidence that levels have fallen below what's considered healthy, and that black and Hispanic children are at particularly high risk.

Both the optimal amount of vitamin D supplementation and the healthy blood level of vitamin D are under heated debate in the medical community. Currently, the American Academy of Pediatrics recommends children should have vitamin D levels of at least 50 nmol/L (20 ng/ml). However, other studies in adults suggest that vitamin D levels should be at least 75 nmol/L (30 ng/ml), and possibly 100 nmol/L (40 ng/ml), to lower the risk of heart disease and specific cancers.

Mansbach and collaborators from the University of Colorado Denver and Massachusetts General Hospital used data from the National Health and Nutrition Examination Survey (NHANES) to look at vitamin D levels in a nationally representative sample of roughly 5,000 children from 2001-2006. Extrapolating to the entire U.S. population, their analysis suggests that roughly 20 percent of all children fell below the recommended 50 nmol/L. Moreover, more than two-thirds of all children had levels below 75 nmol/L, including 80 percent of Hispanic children and 92 percent of non-Hispanic black children.

"If 75 nmol/L or higher is eventually demonstrated to be the healthy normal level of vitamin D, then there is much more vitamin D deficiency in the U.S. than people realize," Mansbach says.

Mansbach and his co-authors suggest that all children take vitamin D supplements, because of the generally low levels that they found and the potential health benefits of boosting vitamin D to normal levels. Vitamin D improves bone health and prevents rickets in children, and recent studies suggest that it also may prevent a host of common childhood illnesses, including respiratory infections, childhood wheezing, and winter-related eczema.

Although sun exposure generates healthy doses of vitamin D, it can also cause skin cancer. Dermatologists and the AAP recommend wearing sunblock, but this actually blocks our skin's ability to make vitamin D. Furthermore, children with more highly pigmented skin require much more sun exposure than fair-skinned children to obtain healthy levels of vitamin D. Vitamin D can also be obtained from certain foods, like liver and fatty fish, but almost all children in the U.S. don't consume these foods in high enough quantities to match the vitamin D that could be provided by summer sunshine or vitamin D supplements.

In the study, children taking multi-vitamins that included vitamin D had higher levels overall, but this accounted for less than half of all children. Mansbach recommends that all children take vitamin D supplements, especially those living in high latitudes, where the sun is scarce in the wintertime.

"We need to perform randomized controlled trials to understand if vitamin D actually improves these wide-ranging health outcomes," Mansbach says. "At present, however, there are a lot of studies demonstrating associations between low levels of vitamin D and poor health. Therefore, we believe many U.S. children would likely benefit from more vitamin D."

Carbon nanotubes affect lining of the lungs

Study shows how carbon nanotubes (CNT)  can affect lining of the lungs (the CNT dose makes the posion)

Carbon nanotubes are being considered for use in everything from sports equipment to medical applications, but a great deal remains unknown about whether these materials cause respiratory or other health problems. Now a collaborative study from North Carolina State University, The Hamner Institutes for Health Sciences, and the National Institute of Environmental Health Sciences shows that inhaling these nanotubes can affect the outer lining of the lung, though the effects of long-term exposure remain unclear.

Using mice in an animal model study, the researchers set out to determine what happens when multi-walled carbon nanotubes are inhaled. Specifically, researchers wanted to determine whether the nanotubes would be able to reach the pleura, which is the tissue that lines the outside of the lungs and is affected by exposure to certain types of asbestos fibers which cause the cancer mesothelioma. The researchers used inhalation exposure and found that inhaled nanotubes do reach the pleura and cause health effects.

Short-term studies described in the paper do not allow conclusions about long-term responses such as cancer. However, the inhaled nanotubes "clearly reach the target tissue for mesothelioma and cause a unique pathologic reaction on the surface of the pleura, and caused fibrosis," says Dr. James Bonner, associate professor of environmental and molecular toxicology at NC State and senior author of the study. The "unique reaction" began within one day of inhalation of the nanotubes, when clusters of immune cells (lymphocytes and monocytes) began collecting on the surface of the pleura. Localized fibrosis, or scarring on parts of the pleural surface that is also found with asbestos exposure, began two weeks after inhalation.

The study showed the immune response and fibrosis disappeared within three months of exposure. However, this study used only a single exposure to the nanotubes. "It remains unclear whether the pleura could recover from chronic, or repeated, exposures," Bonner says. "More work needs to be done in that area and it is completely unknown at this point whether inhaled carbon nanotubes will prove to be carcinogenic in the lungs or in the pleural lining."

The mice received a single inhalation exposure of six hours as part of the study, and the effects on the pleura were only evident at the highest dose used by the researchers – 30 milligrams per cubic meter (mg/m3). The researchers found no health effects in the mice exposed to the lower dose of one mg/m3.

Genes Determine Susceptibility to H5N1 Avian Influenza A Virus Infection

A new study found genetic variations in mice affect their susceptibility to and severity of H5N1 avian influenza A virus infection suggesting that humans who contract the virus may be genetically predisposed. The researchers from St. Jude Children's Research Hospital, VA Medical Center and MidSouth Center for Biodefense and Security, and the University of Tennessee Health Science Center, Memphis, Tennessee report their findings in the October 2009 issue of the Journal of Virology. Over the last 10 years, highly pathogenic H5N1 avian influenza A has spread from Southeast Asia into Europe and Africa killing millions of chickens and ducks along the way. It has also infected tigers, cats, dogs and humans often resulting in death.

Despite the countless cases reported in birds, the number of human cases remains few and of those few more than 90% occurred in genetically related family members indicating a possible genetic correlation. In the study researchers conducted genome-wide linkage analysis to identify chromosomes that contribute to varying susceptibility to H5N1 in two inbred strains of mice challenged with a lethal dose of a highly pathogenic H5N1 virus. Results revealed five quantitative trait loci for influenza virus resistance located on multiple chromosomes also associated with H5N1 resistance.

Additionally, a number of candidate susceptibility genes were identified, one of which affected virus titers 7 days following infection. "An important and novel finding of this study is that H5N1-induced pathology is greatly affected by genetic polymorphisms in the genome of the infected host," say the researchers. "We have also found that, at least in mice, H5N1 pathogenesis is a complex genetic trait with multiple genes affecting disease outcome."

Hydrated Fullerenes and Carbon Nanotubes positively affect plant & animal growth

Researchers  at the Institute for Physiological Active Compounds in Kharkiv Ukraine have in the past two years shown that hydrated fullerenes (C60) HYFNs have a positive effect on both plant and animal growth. One day old chicks, who  are feed water with nano levels of fullerenes are healthier, weigh more and don't need antibiotics. (Hydrated Fullerenes have anti-viral and anti-bacterial properties)

Similarly wheat seedlings dipped in water laced with hydrated fullerenes, grow faster and produce healthier and more tolerant wheat plants.

Now we see similar results with carbon nanotubes. (BTW it may all come down to price--Fullerenes are less expensive to produce then CNT and safer and less expesive then antibiotics. A Fullerene Water Solution (FWS) drink is about to be launched in the Ukrainian market-a world first)

Scientists in Arkansas are reporting that carbon nanotubes (CNTs) could have similar beneficial effects in agriculture. Their study, found that tomato seeds exposed to CNTs germinated faster and grew into larger, heavier seedlings than other seeds. That growth-enhancing effect could be a boon for biomass production for plant-based biofuels and other agricultural products, they suggest.

Mariya Khodakovskaya, Alexandru Biris, and colleagues note that considerable scientific research is underway to use nanoparticles — wisps 1/50,000th the width of a human hair — in agriculture. The goals of "nano-agriculture" include improving the productivity of plants for food, fuel, and other uses.

The scientists report the first evidence that CNTs penetrate the hard outer coating of seeds, and have beneficial effects. Nanotube-exposed seeds sprouted up to two times faster than control seeds and the seedlings weighed more than twice as much as the untreated plants. Those effects may occur because nanotubes penetrate the seed coat and boost water uptake, the researchers state. "This observed positive effect of CNTs on the seed germination could have significant economic importance for agriculture, horticulture, and the energy sector, such as for production of biofuels," they add.

Glacial melting releases pollutants in the environment

Those pristine-looking Alpine glaciers now melting as global warming sets in may explain the mysterious increase in persistent organic pollutants in sediment from certain lakes since the 1990s, despite decreased use of those compounds in pesticides, electric equipment, paints and other products. That's the conclusion of a new study, scheduled for the Nov. 1 issue of ACS' Environmental Science & Technology, a semi-monthly journal. In the study, Christian Bogdal and colleagues focused on organic pollutants in sediment from a model body of water –– glacier-fed Lake Oberaar in the Bernese Alps, Switzerland –– testing for the persistent organic pollutants, including dioxins, PCBs, organochlorine pesticides and synthetic musk fragrances. They found that while contamination decreased to low levels in the 1980s and 1990s due to tougher regulations and improvements in products, since the late 1990s flow of all of these pollutants into the lake has increased sharply. Currently, the flow of organochlorines into the lake is similar to or even higher than in the 1960s and 1970s, the report states.

 

The study attributed the most recent spike in the flow of pollutants into Lake Oberaar to the accelerated release of organic chemicals from melting Alpine glaciers, where contaminants were deposited earlier and preserved over decades. "Considering ongoing global warming and accelerated massive glacial melting predicted for the future, our study indicates the potential for environmental impacts due to pollutants delivered into pristine mountainous areas," Bogdal said.

Innovation is alive and well in Ukraine

When it comes to Ukraine, political bickering, the economic crisis, corruption and scandals and other negative events such as Chornobyl tend to dominate the international headlines.  But, after my most recent two week trip to Ukraine, I can confidently report that despite all of these negative perceptions that make the public spotlight, on the positive side of the ledger, innovation, discovery and creativity is alive and flourishing in Ukraine. In some cases, it can be ranked as world-class. After the fall of the Soviet Union, one of the few positive things that Ukraine inherited was over 1400 academic research institutes and some of the best theoretical scientists in the world  Many of these institutes were classified as secret military establishments  working on weapons of mass destruction. Now this science know-how is being converted to peaceful civilian applications (see www.stcu.int)

 

 

I presented a paper on business opportunities at the nexus of aerospace and aviation at the Canada Ukraine Aerospace and Aviation Business Summit in Kyiv on Sept 28 to Sept 30, 2009, which was sponsored by the Canadian Space Agency, The National Research Council of Canada, The National Ukrainian Space Agency and the Science and Technology Center of Ukraine. Participants included many noteworthy Canadian companies and institutions such as Bombardier, MacDonald Dettwiller and Associates (MDA),  L-3 WESCAM; MDS Corporation,  Novatel, Aerospace Industries Association of Canada, the Canadian Space Agency and the National Research Council of Canada and others.

 

Sun Spring

While a number of Canadian and Ukrainian innovations and collaborative projects were discussed, one project that was proposed by the Ukrainian Space Agency caught my eye. Code named Sun Spring, it extends the observation that every child knows, who is playing with a magnifying glass. Children quickly learn that they can easily focus the sun’s rays with a magnifying glass to burn or heat up a point source on the ground. This same macro effect occurs in space but at orders of magnitude larger. As the sun shines on the earth, the atmosphere which is about 100 miles in circumference, concentrates sunlight just like a magnifying glass does toward a theoretical point source along a spectrum line at a distance from 0.6 to 1.2 million kms from the center of the earth and opposite to the sun. This theoretic power point spectrum rotates along with the earth as the globe rotates around the sun. It creates an estimated power source of 5 gigawatts or the equivalent energy savings of 144,000 tons of coal per day.  The Ukrainian Space Agency plans to tap this Solar Spring by initially sending up satellites to measuring these power levels and locations in space and then launch and mount an array of satellites with solar mirrors in synchronous earth orbit, that would reflect this concentrated sunlight back to the earth to the more or less stationary North and South poles. Just like the existing solar farms in Spain and Quebec, these solar farms would be used to heat steam turbines to produce electricity. This 30 year project, which appears to be feasible with existing technology, is now looking for other international collaborators. Other spinoffs could include: anti-asteroid protection; weather & climate control, acceleration of vegitation & plankon growth in oceans, other geoengineering projects,

 

 

As I toured through various research institutes in Kyiv, Dnipropetrovsk, Kharkiv and Lviv, (many of which would have arrested me as a spy, as little as 17 years ago had I stepped foot on their territory),  I was especially excited by two  medical breakthroughs and commercial follow-up products just hitting the Ukrainian market.. Western researchers are only now realizing that a change in western diets over the past 100 years have shifted our intake of omega 3 fatty acids (found in flax and fish oil) versus omega 6 fatty acids  ( found in meat and vegetable oils) from a healthy  to an unhealthy balance. Anthropological evidence suggests that human ancestors maintained a healthier 2:1 omega 6/ omega 3 ratio for much of history (100,000 generations), but in Western countries today the ratio has spiked to as high as 10:1. Since these omega fatty acids can be converted into inflammatory molecules, this dietary change is believed to also disrupt the proper balance of pro- and anti- inflammatory agents in our bodies, resulting in increased systemic inflammation and a higher incidence of problems including asthma, allergies, diabetes, Alzheimer’s, blood clots (thrombosis), cholesteral, and arthritis.  (See “Effect of dietary fatty acids on inflammatory gene expression in healthy humans”, by Kelly L. Ukrainian researchers have been researching these effects for over a decade and are just launching two new natural food supplements to address these autoimmune and anti-inflammatory diseases that plagued both developed and developing nations. This could obsolesce mainstream pharma's strategy whose  business model is to treat the symptoms and not the disease....read Pharmaceuticals have no interest in curing disease.... a long term, sick patient is a better customer then a healthy one. So, when I felt a cold coming on at night in the freezing-cold sleeping car on the train between Kyiv and Lviv, I was curious if I would get my customary annual bout of influenza or not. I remember that I coughed for about 1/2 an hour trying to clear my raspy throat; I then started to coughed up a frothy watery flem, which I had never experienced before, instead of the traditional green or white mucus which I was expecting to see during a cold. And then, all of a sudden, it stopped as quickly as it started and I fell back asleep.
